Velpatasvirandsofosbuvirarenewdrugsprescribedinacombinedpharmaceuticaldosageformthat poseanewchallengefor thetreatmentofchronichepatitisC. Inthiswork, acomparativeevaluation of theclassicalmathematicalmodel, simultaneousequations, andtheadvancedmathematicalmodel, partial leastsquares, for thespectrophotometricquantitativeanalysisofvelpatasvirandsofosbuvir in bulkpowderandinthenewcombinedpharmaceuticaldosageformwaspresented.Themathematical simultaneousequationmethodwasusedtoresolve theoverlapbetweenvelpatasvirandsofosbuvir. Theabsorbanceandabsorbativityvaluesat255and244.8wereusedtoconstruct twomathematical equationsrequiredforspectrophotometricquantitativeanalysisof thedrugsunderstudy.Partial least squares,anadvancedmathematicaltooldealingwiththefullspectraldataofvelpatasvirandsofosbuvir, wasalsointroduced.Anexperimentaldesignforthecalibrationsetsandvalidationsetsforthebinary mixtureof thedrugsunderstudywerecreated.Themodelwasoptimizedbasedonafive-level, two factorexperimentaldesign.Pre-processingofthespectraldatawasappliedandresultedintheexclusion ofthespectralregionfrom200to230nmduetonoise.Thedescribedmethodsweresuccessfullyapplied tothespectrophotometricquantitativeanalysisofvelpatasvirandsofosbuvirinEpclusa tablets.
