2022 Analysisoftheternaryantiretroviraltherapydolutegravir, lamivudine andabacavirusingUVspectrophotometryandchemometrictools Spectrochimica Acta Part A: Molecular and Biomolecular Spectroscopy
Herein, asimplespectrophotometricmethodcoupledwithchemometric techniques i.e. partial least square(PLS)andgeneticalgorithm(GA)wereutilizedforthesimultaneousdeterminationof thevital ternaryantiretroviral therapydolutegravir(DTG), lamivudine(LMV), andabacavir(ACV) intheircom bineddosageform.Calibration(25samples)andvalidation(13samples)setswerepreparedforthese drugsatdifferentconcentrationsviaimplementingpartialfactorialexperimentaldesigns.Thezeroorder UVspectraofcalibrationandvalidationsetsweremeasuredandthensubjectedforfurtherchemometric analysis.Partialleastsquareswith/withoutvariableselectionproceduresi.e.geneticalgorithm(GA)were utilizedtountangletheUVspectraloverlappingofthesemixtures.Cross-validationandexternalvalida tionmethodswereappliedtocomparetheperformanceof thesechemometrictechniques intermsof accuracyandpredictiveabilities. Itwas foundthat six latentvariableswereoptimumformodelling DTG, four latentvariables formodellingLMVandthreelatentvariables formodellingACV.Although, goodrecoverieswithpromptpredictiveabilitywereattainedbythesePLS,GA-PLSshowedbetteranalyt icalperformanceowingtoitscapabilitytoremoveredundantvariables i.e. thenumberofabsorbance variableshavebeenreduced toabout 21–29%. Theproposedchemometricmethods canbe reliably appliedforsimultaneousdeterminationofDTG, LMV, andACVintheir laboratorypreparedmixtures andpharmaceuticalpreparationposingthesechemometricmethodsasworthyandsubstantialanalytical tools in in-process testing and quality control analysis of many antiretroviral pharmaceutical preparations.
